The overall objective of the Molecular Genetics Core is to provide high quality, low cost DNA sequencing, microsatellite genotyping and a variety of SNP genotyping and gene expression analysis technologies for investigators. We offer high quality, high throughput DNA sequencing, microsatellite and SNP (single nucleotide polymorphism) genotyping using the Applied Biosystems 3730 DNA Analyzer. Microsatellite and multiplex SNP genotyping is performed using our in-house Applied Biosystems MD10 LMS microsatellite screening set, the investigator's custom microsatellite primer sets or the Sequenom MassARRAY, allowing simultaneous genotyping of up to 25 custom SNP's in a single multiplex reaction. In addition, we will assist investigators in designing a genome-wide or candidate locus genotyping project using microsatellites or SNP's. Specifically, we offer expertise in conducting a genetic linkage/association study from evaluating the power of the subject family or cohort to calculation of the linkage score. The Core Facility offers real time (quantitative) PCR for expression analysis and individual SNP genotyping using our Applied Biosystems 7900HT Sequence Detection System. This approach is suitable for analyzing a small number of SNP's in a large number of subjects. We also offer genome-wide SNP array genotyping for linkage analysis with the Affymetrix 10K and 100K GeneChip Human Mapping system as well as gene expression array analysis using the Affymetrix GeneChip microarray expression system. In addition, we have purchased an Illumina BeadStation 600 as a new and even lower cost technology for expression and SNP array analyses. The Molecular Genetics Core Facility is utilized by investigators conducting both clinical and basic research and we will continue to strive to provide investigators with state-of-the-art technology.
